银杏叶提取物对帕金森病大鼠黑质细胞的保护作用 被引量:3

摘要 目的 观察细胞凋亡在帕金森病发病过程中的作用 ,探讨银杏叶提取物用于帕金森病防治的可能性。方法 通过 6 OHDA脑立体定向注射术建立大鼠帕金森病模型 ,采用生化、TUNEL、电镜的方法观察帕金森病大鼠模型制作后 1、7、14、2 1d黑质细胞自由基 (丙二醛、过氧化物歧化酶 )、细胞凋亡数变化及超微结构的改变 ,同时观察使用银杏叶提取物 (EGb组 )治疗对上述指标的影响。结果 在大鼠帕金森病发病过程中PD组、EGb组大鼠黑质丙二醛 (MDA)、过氧化物歧化酶 (SOD)、细胞凋亡数随时间增加而升高 ,PD组大鼠各指标均高于EGb组 (P <0 .0 5 ) ,PD组黑质细胞超微结构改变符合典型的凋亡改变。结论 细胞凋亡参与了帕金森病发病 。 Purpose: To investigate the effects of apoptosis during the pathogenetic process of parkinson disease (PD) and explore the possibility of ginkgo biloba extract (EGb) to prevent and cure PD. Methods: At 1,3,5,7,14,21 d after 6-OHDA injecting stereotaxically to right side of the mesencephic ventral tegmental area (VTA) and substantia nigra pars compacta (SNc) of rats, we used biochemical, immunocytochemical, TUNEL and electronicmicroscopic techniques to observe free radicals (MDA, SOD), apoptosis, and the ultrastructure of substantia nigra in rats in the course of the formation of PD models of rats (PD group). In the same way the indexes of the group treated by EGb(EGb group) and the contral group were observed. Results: In the PD and EGb groups, free radicals (MDA, SOD), the numbers of apoptosis and increased gradually, but the indexes of the PD group surpassed those of the EGb group and the control group (P< 0.05). In the PD group, the ultrastructure of substantic nigra exhibited morphological characteristics of apoptosis from 1 d to 21 d. Conclusions: Apoptosis contributed to the pathagenesis of PD in rats. EGb may mitigate the injury of substantia nigra evidently during the pathogenetic process of PD in rats.
