摘要
目的 揭示连环蛋白与肝癌的发生发展的关系。方法 应用RT -PCT和免疫组织化学观察细胞黏附与信号转导分子 β 和 p12 0 -连环蛋白在正常肝组织、肝癌旁结节性肝硬化组织和肝癌组织中 p12 0 ctnmRNA的蛋白质表达情况及其与β 连环蛋白的关系。 结果 所有肝组织中均表达p12 0 ctnmRNA同工蛋白 1A和 3A。 2例正常肝组织中 β 和p12 0 连环蛋白分子表现为胞膜表达而胞质无表达 ;17例癌旁结节性肝硬化组织表现为胞膜和胞质均有表达 ,以胞质为主 ,且胞膜表达有所增强。 17例HCC组织中表现为胞膜表达明显减弱或消失 ,而胞质表达则增强。多数细胞胞膜表达呈现不连续性。结论 肝组织中可检测到p12 0 ctn同工蛋白 1A和 3AmRNA。此外 ,正常肝组织中 β 和 p12 0 连环蛋白分布在胞膜 ,对维持正常的细胞黏附和信号转导起重要作用 ;结节性肝硬化肝组织中此 2种分子出现了分布变化 ,而肝癌细胞中此 2种分子发生了明显的转位现象 。
Purpose: In order to investigate the relationship between catenins and the mechanism of tumorigenesis, development and transference of human hepatocellular carcinoma. Methods: RT - PCR methods was used to detect p 120ctn isoform 1A and 3A mRNA expression in normal hepatic tissues, hepatocirrhosis tissues and hepatocellular carcinoma tissues. Also we examined p120ctn protein and β - catenin expression in the tissues mentioned above by immunostaining of the specimens. Results: All the tissues expressed the p120ctn isoform 1A and 3A mRNA. In 2 normal hepatic tissues, the molecules expressed at cell membrane, and they were negative in cytosol; While in 17 hepatocirrhosis tissues, the molecules expressed both at the membrane and in cytosol(in the large scale), and membrane expression were increased. In 17 carcinoma specimens, the molecules expressed maily in cytosol, membrane staining reduced significantly or disappeared, also stained cells were uneven. Conclusions: p120ctn isoform 1A and 3A can be detected in liver tissuses. The molecules can keep and 3A can be detected in liver tissuses. The molecules can keep normal adhesion and signaling of the cell in normal hepatic tissues; While it appeared that in hepatocirrhosis which was pre-cancerized like these, the molecules had translocated and the liver underwent compensation and in the progress to cancer; The molecules in hepatocellular carcinoma tissues had translocalized or relocalized, it suggests may be the cancer cells lost normal adhession and signal transduction function.
