摘要
为探讨Ca2 + 激活猪凝血酶原的最适工作浓度和激活时间 ,以新鲜猪血为材料 ,采用HAc等电点沉淀法获取凝血酶原后 ,添加 0 .2 5mol/LCaCl2 作为凝血酶原激活剂 ,使Ca2 + 达到不同终浓度 ,并经过不同激活时间进行对比试验。结果显示以Ca2 + 单一因子激活猪凝血酶原的最适CaCl2终浓度为 0 .0 5mol/L ;最适激活时间为 1.5h。使Ca2 + 激活猪凝血酶原的实验条件得以确立。
In order to approach the optimal working concentration and activating time for porcine prothrombin activation with Ca 2+ , firstly porcin prothrombin was isolated using the method of HAc isoelectric fractionation from fresh porcine blood, then 0.25mol/LCaCl 2 as activator was added in the prothrombin solution. Various final Ca 2+ concentrations and different activating time were tested, and the results showed that with Ca 2+ as single factor to activate porcine prothrombin, the optimal final concentration of CaCl 2 was 0.05mol/L;the optimum activating time was 1.5 hours.The experimental condition for Ca 2+ activating porcin prothrombin was established.
