摘要
本文采用硫酸铵盐析、SephedexG 15 0柱层析从蜂王浆中分离出了超氧化物歧化酶 (SOD)。经聚丙烯酰胺凝胶电泳显示单一蛋白区带。此酶比活力为 1887.4U/mg ,纯化倍数为 5 2 4.3,回收率为 12 .47% ,最适pH 8.3 ,最适温度为 2 5℃。Mn2 + ,Cu2 + ,Ba2 + ,Mg2 + 对此酶有激活作用 ,Zn2 + 则具有抑制作用。过氧化氢和乙醇 氯仿实验表明 :蜂王浆中的SOD属Cu/Zn
Copper Zinc superoxide dismutase was purified from royal jelly by precipitation with ammonium sulfate and Sephadex G150 in 12.47% yield by a purification factor of 524.3. The specific activity of SOD was estimated as 1887.4U/mg protein. The purified enzyme showed a single band on PAGE. The results from treatment with H 2O 2 or ethanol/chloroform,indicated that the enzyme was Cu/Zn SOD. Its optimum pH was 8.3 and optimum temperature,25℃.Mn 2+ ,Cu 2+ ,Ba 2+ and Mg 2+ activate the enzyme and Zn 2+ inhibits the enzyme.
