摘要
研究了分光光度法测定对羟苯基丙酮酸 ;实验表明在碱性介质中 ,对羟苯基丙酮酸的λmax=329nm,其含量在0.001~2mg/L范围内与吸光度值呈良好的线性关系 ,摩尔吸光系数ε为5.26×104L·mol-1·cm-1,方法检出限为0.0078mg/L(以3σ计 ) ;该法操作简单、快速 ,无需昂贵的仪器 ,数据重现性好 ,其最大的优点是可以有效地消除同类物质如对羟苯基丙氨酸、对羟苯基丙乳酸和对羟苯基丙酸等的干扰 ,是测定微量对羟苯基丙酮酸的特效方法 ;该法应用于尿液中对羟苯基丙酮酸的测定 。
A new method for the spectrophotometric determination of p _ hydroxyphenyl pyruvic acid in urine is described. In an alkaline medium content of p _ hydroxyphenyl pyruvic acid has a good linear relationship with absorbance at λmax of 329 nm in the range of 0.001~2 mg/L. The value of molar absorptivity(ε) of the complex of p _ hydroxyphenyl pyruvic acid is 5.26×104 L·mol-1·cm-1. The detection limit of the method is 0.007 8 mg/L(3σ), and the RSD for twelve determination of 1.0 mg/L of p _ hydroxyphenyl pyruvic acid is 3.4%. The method is found to be simple, rapid, repeatable and free from interference of p _ hydroxyphenyl lactamic acid, p _ hydroxyphenyl propyl lactic acid and p _ hydroxyphenyl propionic acid. It was applied to the determination of p _ hydroxyphenyl pyruvic acid in urine with satisfactory results.
