胰岛素样生长因子Ⅰ类受体反义核酸对脐血B淋巴细胞功能的调节 被引量:3

Effects of insulin-like growth factor type Ⅰ receptor antisense nucleotide on cord blood B lymphocyte functions

摘要 目的 探讨胰岛素样生长因子Ⅰ类受体 (IGF ⅠR)与脐血B淋巴细胞增殖以及IgM合成的关系。方法 通过反义核酸阻抑脐血B淋巴细胞IGF ⅠR基因表达水平 ,采用同位素掺入法观察B淋巴细胞的增殖功能 ,利用酶联免疫吸附 (ELISA)双抗夹心法检测IgM含量 ,通过逆转录聚合酶链反应技术检测IGF ⅠR基因表达。结果 经反义寡核苷酸 (ASON)阻抑后 ,脐血B淋巴细胞IGF ⅠRmRNA的表达水平明显降低 ,降低幅度波动于 15 %~ 4 9%;同时B淋巴细胞的增殖功能较单纯美洲商陆 (PWM)刺激下降 5 4 %;细胞活化第 5天IgM的生成量明显降低。结论 IGF ⅠR参与脐血B淋巴细胞增殖、分化以及IgM的产生过程 ,提示IGF ⅠR与IGF Ⅰ结合发挥的生物活性在维持新生儿时期体液免疫水平方面起着重要的促进作用。 Objective To study the relation between insulin-like growth factor type Ⅰ receptor (IGF-ⅠR) and cord blood B lymphocyte proliferation and immunoglobulin M (IgM) production. Methods Antisense oligonucleotide was used to inhibit the expression of IGF-ⅠR mRNA of cord blood B lymphocyte. Lymphocyte proliferation response was measured by 3H-thymidine uptake, and the mRNA expression of IGF-ⅠR was detected by using reverse transcriptase polymerase chain reaction. IgM production was measured by enzyme linked immunosorbent assay (ELISA). Results A 15%~49% reduction in IGF-ⅠR mRNA of B lymphocyte following exposure to antisense oligonucleotide was observed. The proliferative response of B cell to mitogen was significantly reduced while the mRNA level of IGF-ⅠR became lower, and the amplitude of reduction was 54%. The production of IgM was also markedly decreased. Conclusions IGF-ⅠR was involved in the process of cord blood B lymphocyte proliferation and the production of IgM. IGF-ⅠR may play an important role in maintaining humoral immune functions in newborn.
