摘要
目的探讨良性胆管狭窄形成机制。方法通过制作犬胆管损伤修复模型,术后采用免疫组化 SP 法对巨噬细胞(Mφ)、TGFβ_1及α-SMA 在胆道愈合过程不同时期组织中的表达和分布进行研究。结果 Mφ、TGF-β_1及α-SMA 愈合过程中表达较强,且持续较长时间。Mφ阳性表达于粘膜固有层;TGF-β_1主要表达于肉芽组织、成纤维细胞、Mφ及血管内皮细胞。α-SMA 表达于肌纤维细胞胞浆。结论 Mφ、TGF-β_1及α-SMA 高表达是造成胆道愈合过程成纤维细胞增殖旺盛、细胞外基质过度沉积、胆道瘢痕性挛缩的重要因素。
Objective To explore the mechanism of benign biliary stricture.Methods A model of trauma of bile duct was established in dogs and then repaired.The expression and distribution of MΦ,TGF-β_1 and α-SMA in tissues at different time phases of the healing process were studied after the operation with immunohistochemical SP staining.Results High expression of MΦ,TGF-β_1 and α- SMA were observed in the healing process and lasted for a long time.MΦ staining was found in the in- trinsic layer under mucosa,TGF-β_1 staining in the granulation tissue,fibroblasts and endothelial cells of blood vessels while α-SMA staining in the myofibroblasts.Conclusions The high expression of MΦ,TGF-β_1 and α-SMA is closely related to active proliferation of fibroblasts,extracellular matrix overdeposition and scar contracture of bile duct.
