人乳头瘤病毒16、18型分子流行病学及新株筛查 被引量:6

Molecular epidemiological study on sex transmitting HPV16, 18 and the screening of a new HPV strain

摘要 目的 建立浙江省绍兴地区性传播人乳头瘤病毒 (HPV) 16、18型的分子流行病学资料 ,并寻找本地区HPV流行变异株。方法 采用聚合酶链反应 (PCR)扩增HPV公共型、HPV16型和HPV18型的特异性片段并对这些片段进行克隆和测序分析。结果 本地区HPV16和HPV18型感染率占总HPV中的 10 .5 % ;HPV16型的感染率高于HPV18型 ,差异有显著性 (P <0 .0 5 ) ;女性中有 2 6例 (9.4% )为无症状感染 ,其中 15 .4%是与HPV16、18型相关 ;2 0~ 30岁年龄段组感染率最高 ;女性感染率高于男性。利用公共型HPV引物 (L1区 )筛选 ,获得 1株HPV变异株 ,其序列经国际联网BLAST检索 ,证实其氨基酸序列与已知HPV间有 2 5 %的差异。结论  1.本地区HPV16、18型感染率较高 ;2 .在本地区筛查到我们尚未见报道且变异大的HPV株 。 Objective To obtain the molecular epidemiological data of Human Papilloma virus(HPV) 16 and HPV 18 and screen new HPV strains. Methods Common type HPV, HPV16 and HPV18 were amplified by PCR and then cloned and sequenced. Results The infection rates of HPV16、 HPV18 accounted for 10.51% of total HPV infection rate in the area. The infection rate of HPV16 was higher than that of HPV18 ( P <0.05). 26 female carriers were asymptomic and 15.38% of them were infected with HPV16,18. A new subtype of HPV was found by using common HPV primers in this study. 25% sequence of amino acid of the new subtype HPV were different from that of established strains in geneBank. It showed that there existed a new strain of HPV in the area. Conclusions 1. The infection rates of HPV16,18 in local area are higher than those of other reports. 2. New strain has been found in the area, suggesting that unknown strain of HPV is spreading in local area.
