摘要
进行了蜜环菌深层发酵产胞外多糖的培养基的研究。研究结果表明 :以豆粕粉作氮源时 ,菌丝得率最高 ,以麸皮作氮源时 ,多糖产量最高 ;红薯粉为蜜环菌菌丝生长和多糖产量的最适碳源。最适培养基配方为 :红薯粉 3 % ,葡萄糖 1 % ,豆粕粉 1 5 % ,KH2 PO40 1 5 %。机械搅拌对菌丝的生长和多糖的产生都不利 ;接种后当即上摇床比接种后静置一段时间再上摇床的菌丝干重及多糖含量都多 ;培养基中加 1 %的乙醇对菌丝的生长和多糖的产生都不利 ;60 0ml发酵试验中 ,发酵液多糖含量可达 0 485mg/ml,菌丝干重可达2 0 8g/L。当发酵液pH降为 5 0左右 ,颜色开始变为棕褐色 ,菌丝亮光由强变弱时 ,即可终止发酵。
Production medium of exopolysaccharides from armillaria mellea was investigation.The results showed that:Soybean cake powder is the optimal nitrogen source for strain growth,and wheat skin is the optimal nitrogen source for strain to produce polysaccharide.Sweet potato powder iss the optimal carbon source for fermentation of this fungus.The optimal medium composition is:3.0% sweet potato powder,1.0% glucose,1.5% soybean cake powder,and 0.15%KH 2PO 4.mechanical agitation was harmful to strain growth and polysaccharide production.If it was cultured with agitation after being inoculumed,this fungus would produce more exopolysaccharide.When 1.0% alcohol was added to the mdeium,the strain grew worst and exopolysaccharide produced least.The fermentation in 600ml medium showed that 0.485mg/ml exopolysaccharides were contained in fermentation broth,and dry biomass could attain 20.8 g/L.When the strain begins to become colorless and the pH decreases to about 5.0,the fermentation could be stopped.
