摘要
目的 :研究缺血预处理对沙土鼠前脑缺血后海马CA1区延迟性神经元死亡的影响。方法 :沙土鼠前脑缺血再灌注模型 :阻断双侧颈总动脉 5min ,然后松开 ;沙土鼠缺血预处理模型 :在 5min缺血前 2d行 1或 2次 2min缺血。 30只沙土鼠随机分为假手术组 (Sh组 ) ,缺血组 (Is组 ) ,预处理组 (Po组 ) ,缺血预处理 1组 (Ip1组 ) ,缺血预处理 2组 (Ip2组 )。末次缺血后第7d用组织学检查海马CA1区存活的锥体细胞数。结果 :缺血预处理能明显减少沙土鼠前脑缺血后海马CA1区锥体细胞死亡的数目 ,且 2次缺血预处理的保护效果优于 1次缺血预处理。结论
Objective:To investigate the effecs of ischemic preconditioning on hippocampal CA1 delayed neuronal death in gerbils after forebrain ischemia.Methods:The forebrain ischemia was produced in gerbils by occlusion of both common carotids for 5 min,which consistently resulted in delayed neuronal death(DND) in the CA1 region of the hippocampus.30 gerbils were divided into 5 groups:Sham-operated group(group Sh),Ischemic group (group Is),preconditioning only group (group Po),ischemic preconditioning group 1 (group Ip1,single 2-min ischemic treatment,2d before 5 min ischemia),ischemic preconditioning group 2 (group Ip2,double 2-min ischemic treatments at 1 day intervals, 2d before 5 min ischemia).Histological evaluation was used to assess hippocampal ischemic reperfusion injury.Results:Ischemic preconditioning appeared to provide striking histological protection at CA1 neurons.The percentage of survived neurons in group Ip2 was much higher than that in group Ip1.Conclusion:Ischemic preconditioning can provide histological protection of forebrain ischemia in gerbil.
