摘要 目的 研究胆管癌组织中糖基转移酶mRNA的表达水平 ,及其与胆管癌生物学行为的关系。方法 采用一步法mRNA选择性半定量RT PCR ,检测 15例胆管癌和 15例良性胆道疾病胆管组织中N 乙酰氨基葡萄糖转移酶Ⅴ (GnT Ⅴ )、唾液酸转移酶Ⅰ /Ⅲ (ST 6Gal Ⅰ /ST3Gal Ⅲ )mRNA的表达 ,并研究它们与胆管癌浸润和淋巴结转移的关系。结果 胆管癌组织中ST 3Gal Ⅲ、ST6Gal Ⅰ和GnT Ⅴ表达均增强 (P <0 .0 1)。伴有淋巴结转移的胆管癌 (N1)GnT ⅤmRNA的表达量高于未发生淋巴结转移的胆管癌 (N0 ) (P <0 .0 1) ;发生浸润的胆管癌ST3Gal ⅢmRNA的表达量高于未发生浸润的胆管癌 (P <0 .0 1)。结论 胆管癌组织中的一些糖基转移酶的表达发生了改变 ,与胆管癌的生物学行为密切相关。采用半定量RT PCR检测糖基转移酶的表达 ,将有助于对胆管癌的诊断和预后判断。 Objective To study the expression of glycosyltransferases in tissues of bile duct carcinomas,its correlation with biologic features and clinical significance. Methods The expression of glycosyltransferases mRNA,including sialyltransferases ST3Gal-Ⅲ and ST6Gal-Ⅰ,β_1,6-N-acetylglucosaminyltransferase Ⅴ(GnT-Ⅴ),was detected simultaneously in extracts of the specimens of 15 bile duct carcinomas(BDC) and 15 bile ducts of benign biliary diseases(BBD) by semiquantitative one-step reverse transcription-polymerase chain reaction(RT-PCR) with mRNA selective PCR.Correlation of glycosyltransferases mRNA expression with tumor invasiveness and lymph node metastasis was explored by statistical analysis.Results ST3Gal-Ⅲ,ST6Gal-Ⅰand GnT-Ⅴexpression was all enhanced in BDC(P<0.01).GnT-ⅤmRNA expression was higher in BDC with lymph node metastasis than without metastasis(P<0.01);and expression of sialyltransferase ST3Gal-Ⅲ was increased in BDC with infiltration(P<0.01).Conclusions The expression of some glycosyltransferases is significantly altered in BDC,and they correlate with biologic features of BDC.Semiquantitative RT-PCR determination of specific glycosyltransferases may be helpful for diagnosis and prognosis of BDC.
