摘要
目的 探讨膀胱移行细胞癌组织中MDR 1基因产物P gp表达与凋亡相关蛋白 p5 3、bcl 2的相互关系及其临床意义。方法 采用免疫组化S P法检测 10 7例原发性膀胱移行细胞癌组织中P gp、p5 3、bcl 2蛋白的表达情况。 结果 P gp、p5 3、bcl 2蛋白的阳性表达率分别为 6 3.6 %、72 .9%、5 4.2 % ,三者的过度表达均与膀胱癌的病理分级、临床分期和术后腔内化疗后复发有关 ;P gp表达与 p5 3、bcl 2蛋白的表达密切相关 (P <0 .0 1或P <0 .0 5 )。 结论 P gp、p5 3、bcl 2蛋白的过度表达不仅是判定原发性膀胱移行细胞癌恶性程度和预后的重要指标 ,而且凋亡相关蛋白 p5 3、bcl 2可能参与膀胱移行细胞癌多药耐药的形成。
Objectives To study the clinical significance and relationship between expressions of multi-drug resistance gene(MDR1) product P-glycoprotein(P-gp) and apoptosis associated protein mutant p53,bcl-2 in human primary bladder transitional cell carcinomas(TCCs).Methods 107 human primary bladder TCCs previously untreated were analyzed for expression of P-gp,Mutant p53 and bcl-2 by S-P immunohischemistry(IHC).The mouse′s monoclonal antibodies of JSB1,DO-7 and 100/D5 were used respectively to detect the antigens of P-gp,p53 and bcl-2,respectively.Results P-gp,p53 and bcl-2 were present in 63.6%,72.9% and 54.2% of TCCs.The expressions of P-gp,p53 or bcl-2 were significantly correlated with the grade,stage and recurrence after post-operative intravesical chemotherapy.The expressions of P-gp were strongly related to the expression of mutant P53 and bcl-2(P-gp/p53,P<0.01;P-gp/bcl-2,P<0.05).Conclusions Overexpressions of P-gp,mutant p53 and bcl-2 can be used as the important markers of aggressiveness and prognosis of TCCs.The apoptosis associated mutant p53 protein,bcl-2 protein are probably involved in the formation of multi-drug resistance of TCCs.
