The Correlation of Androgen Receptor and Proliferating Cell Nuclear Antigen to Clinicopathological Feature in Meningiomas

摘要 目的 探讨雄激素受体 (AR)在脑膜瘤组织中的表达及其与肿瘤增殖潜力的关系。方法 采用免疫组化SABC法 ,检测AR、增殖细胞核抗原 (PCNA)在 39例脑膜瘤组织中的表达。结果  5 1.3% ( 2 0 /39)的脑膜瘤组织中AR呈不同程度的表达。良性、非典型性、恶性脑膜瘤中AR表达率分别为 31.6 % ( 6 /19)、5 8.3% ( 7/12 )、87.5 % ( 7/8)。恶性脑膜瘤中AR阳性细胞数显著高于非典型性和良性脑膜瘤 (P <0 .0 5 )。恶性脑膜瘤平均PCNA标记指数 (PCNALI)明显高于非典型性 (P <0 .0 5 )和良性脑膜瘤 (P <0 .0 5 )。AR阳性脑膜瘤的PCNALI高于AR阴性脑膜瘤 (P <0 .0 5 ) ,AR表达与PCNA的表达呈正相关。结论 脑膜瘤AR表达与其病理级别有关 ,AR参与了肿瘤的生长和血管生成 ;检测脑膜瘤中AR表达可间接反映肿瘤的增殖潜力。 Objective To investigate the expression of androgen receptor(AR) in meningiomas and its relation to tumor proliferative potential.Methods Paraffin embedded tumor sections of 39 cases of meningiom were detected with monoclonal antiandrogen receptor antibody and monoclonal PCNA antibody by avidin-biotin complex immunohistochemistry. Results Twenty of the thirty -nineth meningiomas(51.3%) showed positive AR immunoreactivity. The AR positive rates were 31.6%(6/19) in benign meningiomas,58.3%(7/12) in atypical meningiomas,87.5%(7/8) in malignant meningiomas,respectively.Malignant meningiomas had a significantly higher percentage of AR positive cells compared with atypical and benign meningiomas (P<0.05).The mean proliferating cell nuclear antigen labeling index(PCNA LI) was significantly higher in the malignant meningiomas compared with atypical meningiomas (P<0.05) and benign meningiomas (P<0.05). AR positive meningiomas had higher PCNA LI compared with AR negative meningiomas (P<0.05).The expression of AR in tumor tissues was significantly related with PCNA LI.Conclusion AR in the meningiomas correlates with histologic grade and may participate in the growth and angiogenesis of these tumors.The measurement of AR in these tumors may indirectly represent tumor growth potential.
