白细胞介素-11对CITP患者骨髓巨核细胞的影响 被引量:7

摘要 目的 :观察重组人白细胞介素 11(rh IL - 11)对慢性特发性血小板减少性紫癜 (CITP)患者骨髓巨核祖细胞增殖和成熟的影响。方法 :在 2 1例 CITP患者骨髓巨核祖细胞培养体系中加入不同浓度的 rh IL - 11,用血浆凝块法培养 14d后 ,用 ABC试剂盒染色观察集落生长数 ,用 MCDS- 2 0 10骨髓细胞分析系统测定巨核细胞的面积、直径 ;15例健康人作对照组。结果 :CITP组 CFU- MK、BFU- MK总集落数较对照组略低 (P >0 .0 5 ) ,而CITP组巨核细胞面积、直径均低于对照组 (P <0 .0 5 ) ;不同浓度 rh IL- 11在体外对 CITP组及对照组的巨核组细胞集落生成及成熟度均有促进作用 ,呈剂量依赖性 ,以 10 0 μg/ L 作用最佳 ;骨髓涂片巨核细胞数与 CFU- MK及总集落数呈正相关 (P <0 .0 5 ) ,BFU - MK、面积、直径与巨核细胞数无相关性 (P >0 .0 5 )。结论 :CITP患者的骨髓巨核细胞存在成熟障碍。 rh IL - 11对 CITP患者的骨髓巨核祖细胞的增殖与成熟度均有促进作用 ,并呈剂量依赖性。 Objective:To observe the effects of recombinant human interleukin 11 (rhIL 11) on the megakaryocyte (MK) colony growth and maturation in patients with CITP in vitro.Method:Bone marrow (BM) mononuclear cells of twenty one patients were cultured in plasma clot culture systems,to which were added rhIL 11 with different concentrations.After 14 days the MK colonies were stained by ABC Kit and counted.The BM cell analysis system of MCDS 2010 was used to measure the diameters and areas of the positive cells.Result:The number of total MK colonies CFU MK and BFU MK in patients and control group were no significant difference P> 0.05 .Whil the diameters and areas of MK in CITP group were significant lower than those in control group P< 0.05 .rhIL 11 of different concentrations promoted growth in total colonies areas and diameters of MK of patients with CITP and control group.This effect was dose dependent.High concentration (100 ng/ml) was relatively optimal on pronoting proliferation of MK colonies.A Significant positive correlation was found between the numbers of CFU MK,total MK colonies and the number of megakaryocyte on bone marrow smears in patients with CITP.Correlation coefficient was r= 0.718 ,r= 0.719 ,P< 0.05 .Conclusion:There are maturation disfunctions of MK progenitors in patients with CITP.rhIL 11 can promote the proliferation and maturation of MK progenitors in patients with CITP.This effect was dose dependent and no significant difference compared with control group.
