摘要
目的 研究MTT法原代癌细胞药物敏感性试验及影响因素。方法 采用MTT比色法 ,测定 45例 (胃癌 15例 ,肺癌 10例 ,大肠癌 12例 ,乳癌 8例 )体外原代癌细胞对 8种常用化疗药物的敏感性。结果 4种肿瘤细胞对 8种化疗药物的敏感性不同。采用30分钟消化时间、10 0 %~ 6 0 %密度梯度淋巴细胞分离液以及标本离体后即刻实验 ,活细胞百分率为 94 6 5± 2 8 47,与其它实验条件相比差异显著 (P <0 0 1)。结论 MTT法是一种简便、快捷的肿瘤体外药敏试验方法 ,为临床实验个体化化疗提供了实验依据。消化时间、淋巴细胞分离液密度梯度。
Objective To study the anti-cancer drugs' sensitivity in the colormetric assay of cancer cells with MTT and its effective factor.Methods 45 primary cancer cell patients in vitro (15 samples of gastric cancer, 10 samples of lung cancer, 12 samlples of colon cancer, 8 samples of breast cancer) were examined with MTT assay to find out their drugs' sensitivities to the often-used chemotherapy medicine.Results The sensitivity of the 4 kinds of cancer cells to the 8 kinds of chemotherapy medicine is different.Using 30 minute digestion time, 100%~60% concentration of lymphocyte seperating liquid and doing the assay as soon as the sample obtained,the percentige of 94 65±28 47, of the living cell,is significant different with the other experimental condition.Conclusions MTT assay is an easy and quick method for drug-sensitivity test in vitro and provides experimental proof for individual chemotherapy.Digestion time,concentration of lymphocyte seperating flud and the time of sample storager may be important factrs for the results of the assay.
