外源精氨酸摄入对移植瘤生长的抑制作用 被引量:3

摘要 目的:研究精氨酸 (Arginine ,Arg)对移植瘤生长的影响及初步机制。方法 :建立小鼠S180移植瘤模型并胃灌精氨酸 (Arg)、精氨酸 -精氨酸甲酯 (Arg -NAME)及甘氨酸 (Gly) ,检测脾T、B细胞增殖反应程度及肿瘤重量。结果 :Arg补充达 6 .0mg/ (g·d)时 ,T、B细胞增殖反应程度提高至与对照组差异显著 (Arg -Tcell:0 .0 0 5± 0 .0 13,Gly -Tcell:- 0 .0 6 8± 0 .0 2 5 ,P <0 .0 1;Arg -Bcell:0 .0 73± 0 .0 18,Gly -Bcell:- 0 .0 30±0 .0 43,P <0 .0 1) ;Arg -NAME可显著抑制Arg造成的T、B细胞功能的增强 (Arg -Tcell:0 .0 0 5± 0 .0 13,Arg-Bcell:0 .0 73± 0 .0 18;Arg -NAME -Tcell:- 0 .0 2 4± 0 .0 0 7,Arg -NAME -Bcell:- 0 .0 33± 0 .0 10 ,P <0 .0 1) ;Arg补充达 6 .0mg/ (g·d)可产生较Gly组与Arg -NAME组显著较小的瘤重 (0 .78g± 0 .2 1g ,P <0 .0 1)。结论 Objective:To determine the effect of l-arginine on the growth of transplanted tumour and its mechanism.Methods:The rat models of transplanted S180 were established and then different dose of l-arginine,l-arginine-N G-nitro-l-arginine methyl ester (NAME) and glycine were orally instilled.The proliferative levels of T and B cells, the tumor weights were measured.Results:When the supplemented l-arginine was at 6.0 mg/g/day,the improvement of the proliferative levels of T and B cells were significantly different from the control group (Arg-Tcell:0.005±0.013,Gly-Tcell:-0.068±0.025, P <0.01;Arg-Bcell: 0.073 ±0.018,Gly-Bcell:-0.030±0.043, P <0.01);meanwhile the improvement of the fuctions of T and B cells were inhibited by l-arginine-NAME(Arg-Tcell:0.005±0.013,Arg-Bcell:0.073±0.018;Arg-NAME-Tcell:-0.024±0.007,Arg-NAME-Bcell:-0.033±0.010, P <0.01).The tumor weight (0.78g±0.21g, P <0.01) was lighter than those in the control group and group l-arginine-NAME when supplementd l-arginine at 6.0mg/g/day.Conclusions:Supplementing l-arginine can cause the improvement of immunologic function is an important mechanism for the inhibitory effect of l-arginine on tumor.
