摘要
目的 探讨nm 2 3/NDPK癌基因在胰腺癌组织中的表达及其临床意义。方法 采用免疫组化SP法 ,对 40例胰腺癌组织及 10例正常胰腺组织进行nm 2 3/NDPK癌基因检测。结果 40例胰腺癌组织中有 2 6例nm 2 3/NDPK表达阳性 ,占 6 5 % ;10例正常胰腺组织中仅 3例nm 2 3/NDPK表达阳性 ,占 30 % ,两者有显著差异 (P <0 .0 5 )。低分化腺癌nm 2 3/NDPK阳性表达率明显高于高分化胰腺癌 ( 10 / 11,90 .9% ;2 / 8,2 5 % ;P <0 .0 5 )。其nm2 3/NDPK表达阳性与淋巴结转移密切相关 ( 10 / 14 ,71.4% ;6 / 1931.5 % ;P <0 .0 5 )。提示胰腺癌nm 2 3/NDPK表达与淋巴结转移及肿瘤侵袭性呈正相关 ,与癌组织的分化程度呈负相关。结论 nm 2 3/NDPK表达可作为胰腺癌恶性程度及预后不良的生物学指标之一。
Objective To study the expression and clinic significance of nm23 gene (product of nucleoside diphosphate kinase) in the pancreatic adenocarcinoma tissues.Methods S P immunohistochemical method was used to examine the expression of nm23/NDPK in 40 pancreatic adenocarcinoma and 10 normal pancreas tissues.Results It was found that 26 of 40(65%) pancreatic adenocarcinoma showed strong immunoreactivity for NDP kinase,whereas 3 of 10 (30%) normal pancreatic tissues showed weak immunoreactivity (P<0.05).Significant difference was found between the two groups.The nm23/NDPK expression levels in pancreatic adenocarcinoma of lower differentiation were higher than those in pancreatic adenocarcinoma of higher differentiation (10/11,90.9%;2/8 25%;P<0.05).Positive staining was associated with higher incidence of lymph node metastasis (10/14,71.4%) than negative staining (6/19,31.5% P<0.05).These results suggested that nm23/NDPK expression was positively associated with lymph node metastases and aggressiveness,and that nm23/NDPK expression had negative correlation with the extent of histologic differentiation.Conclusion The level of nm23/NDPK can serve as a marker for malignant potentiality and poorer prognosis of pancreatic adenocarcinoma.
