摘要
目的 :建立人子宫内膜癌的原位移植模型 ,观察其生物学特性。方法 :利用子宫内膜癌细胞株AN3CN制备裸鼠皮下移植瘤模型 ,然后 ,将瘤组织条植入裸鼠子宫腔内 ,建立人子宫内膜癌的原位移植模型 ,用组织病理学检查、电镜观察和流式细胞仪DNA含量及染色体核型分析鉴定模型。结果 :原位移植模型的瘤细胞与AN3CN在形态和结构上一致。结论 :小鼠肿瘤的生长和转移完全模拟人子宫内膜癌的临床过程 。
Objective:To establish the orthopotic implantation nude mice model of endometrium carcinoma and to observe its biological features.Methods:At first AN3CN cell line was inoculated subcutaneously into nude mice and grew progressively.The subcutaneous tumor was removed,striped and then implanted into the uterus cavity of nude mice.The model was identified by histological and electronmicroscopic observation.DNA content was analysed by flow cytometry.Results: The implant uterus tumor displayed remarkable invasive growth behavior and extensive distant metastasis after implanting in situ into athymic nude mice.Light and electron microscope and FACS assay revealed that both implant uterus tumor and metastatic tumor showed same morphological and biologic characteristic as original AN3CN cell line.The orthopotic implantation model which exhibit various features seen in clinical endometrium carcinoma:local growth,regional invasion,lymphnode metastases were established successfully. Conclusions:The orthopotic implantation model may be a good tool for research of metastatic mechanism and antitumor therapy.
