钙拮抗剂对成纤维细胞胶原基因的影响

The effect of calcium antagonists on expression of collagen gene in fibroblast

目的 研究钙拮抗剂汉防己甲素 (Tet)、维拉帕米 (Ver)和硝苯啶 (Nif)对 3T3成纤维细胞Ⅰ、Ⅲ和Ⅳ胶原基因表达的影响 ,旨在探讨其抗肝纤维化的机理。方法 应用标记Ⅰ、Ⅲ和Ⅳ型前胶原cDNA探针与RNA斑点印迹杂交技术 ,检测Tet、Ver和Nif与 3T3成纤维细胞共育前后Ⅰ、Ⅲ和Ⅳ型胶原mRNA水平的变化。结果 Tet、Ver和Nif与 3T3成纤维细胞共育后Ⅰ、Ⅲ和Ⅳ型胶原mRNA水平有不同程度降低。结论 钙拮抗剂可以抑制细胞外基质胶原基因的表达。

Aim To study the effects of Tetrandrine(Tet),Verapamil(Ver) and Nifedipine (Nif) on gene expression of collagen types Ⅰ,Ⅲ and Ⅳ in 3T3 fibroblasts,and investigate the mechanism of their effect of anti hepatic fibrosis.Methods The effects of Tet,Ver and Nif on the levels of procollagen types Ⅰ,Ⅲ and Ⅳ mRNA were semi quantified by dot hybridization with procollagen types Ⅰ,Ⅲ and Ⅳ cDNA probes.Results The levels of procollagen types Ⅰ,Ⅲ and Ⅳ mRNA in 3T3 cell groups treated with Tet,Ver and Nif were reduced to different degrees.Conclusion These results sugges that effect involve in inhibition of gene expression of collagen typesⅠ,Ⅲ and Ⅳ.
