化学性肝纤维化中血小板源性生长因子的动态变化 被引量:4

摘要 目的 :研究血浆血小板源性生长因子 (platelet derivedgrowthfactor,PDGF)及病变区PDGFA ,B链表达在化学性肝纤维化不同病理阶段的动态变化。方法 :选用Wistar大白鼠通过皮下注射 4 0 %四氯化碳花生油混合液复制化学性肝纤维化动物模型。采用生物活性方法测定血浆PDGF ,通过肝脏免疫组化染色观察病变区PDGFA ,B链分布变化。结果 :随着肝纤维化病变的加重 ,血浆PDGF逐渐升高。与对照组及相邻病变阶段比较均有显著差异(P <0 .0 1)。通过肝脏免疫组化表明PDGFA ,B链表达早期集中在汇管区间质细胞 ,散在分布于肝窦状隙 ,晚期广泛分布于纤维间隔内。结论 Objective: Our purpose was to study the dynamic alteration of plasma platelet derived growth factor (PDGF) and the expressions of A and B chains of PDGF in the abnormal area of liver in the different pathologic stages of cirrhosis. Methods: We induced the animal models of chemical cirrhosis by hypodermis injection of 40% carbon tetrachloride mixed with peanut oil.The bioactive method was used to detect the amount of plasma PDGF. The changes of distribution of A and B chains of PDGF were observed by using immunohistochemical staining method. Results: The amount of PDGF increased gradually with the development of lesion,which was statistically significant compared with the controls and the neighboring groups .The immunohistochemical staining showed that in the early stage the expressions of A and B chains of PDGF concentrated in the mesenchymal cells in portal area ,scattering in the sinusoid spaces. In the later stage , the expressions of A and B chains of PDGF broadly distributed within the fibrotic septum and mesenchymal cells in portal area. Conclusion: The PDGF plays a promoting role in the progression of chemical liver cirrhosis.
