卡托普利对自发性高血压大鼠心血管重塑的影响

摘要 目的 探讨早期卡托普利对自发性高血压大鼠 (SHR)心血管结构重塑的影响。 方法 雄性 SHR从宫内期起给予卡托普利 (10 0 m g· kg- 1 · d- 1 )至 16周龄 ,未治疗 SHR及正常血压的 WKY为对照 ,3组动物均于16周龄时测血压、体重后处死。检测左心室血管紧张素 含量 ,心肌内冠状动脉周围及间质胶原容积分数 ,主动脉、肾动脉、肠系膜动脉三级分支的血管壁 /腔面积比。 结果 卡托普利明显降低收缩压 ,抑制 SHR的左心室局部血管紧张素 的产生 (2 .4± 0 .3vs 3.0± 0 .4ng/L ,P<0 .0 5 ) ,预防左室 (mg/g 2 .6 1± 0 .2 0 vs 3.5 9± 0 .35 ,P<0 .0 5 )和血管壁肥厚 ,及心肌纤维化 CVF(胶原容积分数 ) 0 .0 7± 0 .0 2 vs 0 .13± 0 .0 3,P<0 .0 5 ,血管周围胶原分数 0 .5 9± 0 .0 7vs 0 .78± 0 .0 5 ,P<0 .0 5。 结论 卡托普利阻止高血压大鼠心血管重塑的发生 ,对预防高血压靶器官损害和改善高血压的长期预后有重要意义。 Objective\ To investigate captopril treatment on the cardiovascular structural remodeling in spontaneously hypertensive rats.\ Methods\ Male SHRs were given captopril(100 mg·kg\+\{-1\}·d\+\{-1\}) from intrauterine period to 16 weeks old.\ Age\|and sex\|matched untreated SHR and normotensive Wistar Kyoto(WKY) rats served as controls.\ After measurement of systolic blood pressure and body weight, rats were killed at 16 weeks old.\ Left ventricles were weighed and then subject to the measurement of angiotensin Ⅱ concentration and collagen volume fraction.\ Vascular medium and lumen ratio from aorta, renal arteries and branch Ⅲ vessels of mesenteric arteries were determined by morphometric method.\ Results\ Captopril markedly reduced systolic blood pressure, prohibited the generation of cardial angiotensin Ⅱ, prevented left ventricular hypertrophy, artery walls thickening and myocardial fibrosis in SHR.\ Conclusion\ Captopril prevented cardiovascular remodeling, demonstrated a clinical implication in the protection of target organs damage and a long\|term favourable prognosis in hypertensive patients.\;
