摘要
目的 :观察新风胶囊对佐剂性关节炎大鼠治疗作用及对滑膜细胞、胸腺淋巴细胞超微结构的作用。方法 :将 6 0只大鼠随机分为正常对照组、模型对照组、新风胶囊组和雷公藤组 ,每组 15只 ,制备完全佐剂并分别向除正常对照组以外的其余动物右后足跖皮内注射 0 .1m l致炎。电镜观察滑膜细胞、胸腺淋巴细胞超微结构变化 (包括线粒体肿胀、空泡变、嵴突病变等 ) ,并计算各组滑膜细胞和胸腺淋巴细胞线粒体的病变率。结果 :新风胶囊组及雷公藤组治疗后鼠右后足跖肿胀度显著低于治疗前 (P均 <0 .0 5 ) ,与模型组相比 ,新风胶囊组和雷公藤组治疗后关节滑膜细胞线粒体的病变率均显著降低 (P均 <0 .0 5 ) ;新风胶囊组胸腺淋巴细胞线粒体病变率显著降低 (P<0 .0 5 ) ,而雷公藤组胸腺细胞线粒体病变率无显著差异 (P>0 .0 5 )。治疗后新风胶囊组滑膜细胞及胸腺淋巴细胞线粒体病变率均显著低于雷公藤组 (P均 <0 .0 5 )。结论 :新风胶囊与雷公藤一样能降低佐剂性关节炎大鼠病变关节的肿胀度 ,新风胶囊改善佐剂性关节炎滑膜病变和对胸腺的保护作用优于雷公藤 ,这可能是其治疗作用的形态学基础。
Objective:To observe the effect of Xinfeng capsule (XFC,新风胶囊)on the ultrastructure of synoviocyte and thymuslymphocyte of the rat model with adjuvantinduced arthritis(AA).Methods:Sixty rats were randomly divided into normal control group,model control group,XFC group and Triperygium Wilfordii Polycoside tablet (TPT,雷公藤多甙片), 15 cases in every group.Complete adjuvant was preparated and 0.1 ml of it was injected into rightposterior subcutaneous part of every animal′s pediphalanx except normal control animals.Electron microscope was used to assess ultrastructural lesion of synoviocyte and thymuslymphocyte (including chondriosome swelling,vacuolation,and ridge pathologic change).The chondriosome pathologic change rate of every group was measured respectively.Results:The swelling degree of rightposterior part of pediphalanx in XFC group and TPT group were lower after treatment than before treatment (all P <0 05).Compared with model control group,the chondriosome pathologic change rate of synoviocyte in XFC and TPT group were lower (all P <0 05),and chondriosome pathologic change rate of thymuslymphecyte in XFC group was lower ( P <0 05),but above parameter in TPT group was not marked variation( P >0 05).Compared with TPT group,chondriosome pathologic change rates of synoviocyte and thymuslymphocyte in XFC group were markedly lower (all P <0 05).Conclusions:In common with TPT,XFC is able to reduce the swelling degree of AA rat,but it is superior to TPT in improving synoviocyte pathologic change and protecting thymus of AA rat.This is likely the morphological basis of XFC′s therapeutic effect.
