摘要
目的 观察CDK4和p16蛋白在鼻咽癌组织中的表达 ,探讨其与鼻咽癌发生发展的关系。方法 应用S P法检测CDK4和p16蛋白在 80例鼻咽癌、30例鼻咽黏膜慢性炎组织中的表达。结果 80例鼻咽癌中CDK4和p16表达率分别为 78 8%和 2 0 0 %。CDK4表达率在鼻咽癌与中重度异型增生黏膜间差异不显著 ,二者与轻度异型增生黏膜相比均差异非常显著 (P <0 0 1)。角化性鳞癌p16的表达率明显高于分化型非角化性癌和未分化癌 (P <0 0 1)。淋巴结转移组中p16表达率明显低于无转移组 (P <0 0 1) ,而CDK4表达率高于无转移组 (P <0 0 5 )。结论 CDK4过表达与p16低表达可能涉及鼻咽癌的发生发展过程 ,CDK4基因的扩增或过表达可能是鼻咽黏膜上皮细胞癌变的早期事件。p16的表达缺失与鼻咽癌的转移、组织学分型有关。
Objective To evaluate the role of CDK4 and p16 protein in nasopharyngeal carcinoma (NPC). Methods S-P immunohistochemical method was used to detect the expression of CDK4 and p16 protein in 80 cases of NPCs and 30 inflammatory nasopharyngeal mucosa. Results Positive rates of CDK4 and p16 expression in NPCs were 78.8% and 20.0% respectively. There was no significant difference in the expression of the above proteins between NPCs and moderate severe dysplasia of nasopharyngeal mucosa. But in the mild dysplasia, the expression of CDK4 and p16 was much higher (P<0.01). Expression of p16 was higher in keratinizing squamous cell carcinoma than that in non keratininzing carcinoma (P<0.01). A significant correlation was found between low expression of p16 and higher metastatic rate in cervical lymph nodes(P<0.01), and between over expression of CDK4 and lower metastatic rate nodes (P<0.05). Conclusions Inactivation of p16 gene and over expression of CDK4 may be involved in the carcinogenesis and development of NPC, and related to histological types and metastasis of the tumor.
