人甲状腺肿瘤促性腺激素释放激素受体的免疫组化

摘要 目的 探讨人甲状腺肿瘤是否存在促性腺激素释放激素受体 (Gn RH- R)及细胞定位 .方法 从我院普通外科收集病例标本 ,分为甲状腺腺瘤组和甲状腺癌组 ,以瘤旁甲状腺及正常甲状腺为对照 ,采用兔抗 Gn RH抗独特型多克隆抗体的免疫组织化学 ABC法 ,确定 Gn RH- R在甲状腺肿瘤中的定位 ,并结合图像分析检测 Gn RH- R在两种肿瘤中相对含量的变化 .结果 甲状腺腺瘤及甲状腺癌细胞均呈较强的Gn RH- R免疫反应阳性 ,阳性物质分布在胞质 ,胞核呈阴性 .甲状腺瘤的阳性信号强于甲状腺癌 .瘤旁甲状腺及正常甲状腺滤泡上皮细胞免疫反应阳性较弱 ,胞质深染 ,呈棕褐色 ,胞核淡染 ,未见有阳性物质分布 .图像分析结果 ,甲状腺瘤组单位面积免疫反应阳性物质的相对含量为 0 .0 74± 0 .0 13,甲状腺癌组为 0 .0 33± 0 .0 11,甲状腺瘤组相对含量显著高于甲状腺癌组 (P<0 .0 5 ) .瘤旁及正常甲状腺组为 0 .0 18± 0 .0 0 7,其相对含量较肿瘤组低 (P<0 .0 5 ) .结论 人甲状腺肿瘤中有Gn RH- R存在 . Gn RH- R含量的多少可能与甲状腺肿瘤的分化程度相关 .我们推测 Gn RH可能参与甲状腺肿瘤发生的调节 . AIM To study presence of gonadotropin releasing hormone receptor (GnRH R) in human thyroid tumor and its cellular location. METHODS Immunohistochemical ABC method and image analysis were used to study the expression and location of Gn RH R in human thyroid adenoma and carcinoma. RESULTS Partial cells of thyroid adenoma and carcinoma showed immunoreactivity. The reactive substances were distributed in cytoplasm of all positive cells with immuno negative nuclei. The staining intensity in thyroid adenoma was stronger than that in thyroid carcinoma. And normal thyroid follicular epithelia cells adjacent to tumor and normal thyroid follicular epithelia cells showed GnRH R immunoreactivity too. The cytoplasm was dark brown while the nuclei showed no staining. Image analysis suggested that relative quantification of GnRH R was 0.074±0.013, in thyroid adenoma and 0.033±0.011 in thyroid carcinoma, which suggested a significant difference ( P <0.05). And the relative amount in normal thyroid follicular epithelia cells adjacent to tumor and normal thyroid follicular epithelia cells was 0.018±0.007, which was lower than that in tumor group ( P <0.05). CONCLUSION There is GnRH R in thyroid tumor indeed. GnRH R may correlate to differentiation degree of thyroid tumor. It suggests that GnRH may take part in the regulation of developing thyroid tumor.
