环孢菌素A对儿童过敏性紫癜T细胞功能状态的干预作用

Role of cyclosporin A in T-cell's function state of children with Henoch-Schonlein purpura

目的 观察环孢菌素A(CsA)对儿童过敏性紫癜 (HSP)T细胞功能状态的干预作用。方法 采用流式细胞技术及ELISA法检测PBMC细胞膜上CD40L表达及PBMC培养上清中细胞因子水平。结果 CsA在体外具有纠正HSP患儿PBMC异常表达CD40L的作用 (P <0 0 5 ) ,并抑制TH2类细胞因子 (IL 4、IL 5、IL 6 )的过度产生 ,使TH1类细胞因子 (IL 2、IFN γ)的释放进一步减少 (P <0 0 5 )。结论 CsA具有显著抑制HSP患儿T细胞功能作用 ,尤其是对T细胞亚群 (TH1)

Objective\ To investigate the roles of cyclosporin A(CsA)in T-cells of Henoch-Schonlein purpura(HSP)patients.Methods\ CD40L expression on peripheral blood mononuclear cells(PBMC)were analyzed by flow cytometry(FACS).Cytokines level in the supernatants of cultured PBMC were detected by ELISA.Results\ Huyperexpression of CD40L and the enhanced T H2 cytokines such as IL 4,IL 5 and IL 6 could be affected by CsA in vitro.The T H1 cytokines were furtherly inhibited.Conclusion\ CsA can significantly inhibit T-cell's function,especially T H1 function.
