血管内皮生长因子、p53蛋白在大肠癌中的表达及其与血管生成的关系

摘要 目的 研究大肠癌中血管内皮生长因子 (VEGF)、p5 3蛋白表达与血管生成的关系以及三者在大肠癌发展中的作用。方法 应用 L SAB免疫组化方法检测 5 2例大肠癌中 MVD、VEGF和 p5 3蛋白的表达。结果 大肠癌中 MVD、VEGF表达与 Dukes分期、淋巴结转移相关 (P<0 .0 5 ) ,而与组织学类型、分化程度无关 (P>0 .0 5 ) ;p5 3蛋白表达与上述各临床病理因素均无关 (P>0 .0 5 ) ;VEGF阳性组的 MVD为 2 4.44± 10 .34 ,明显高于 VEGF阴性组的 15 .88± 6 .41(P<0 .0 1) ,而 p5 3蛋白表达与 VEGF表达或 MVD无相关性 (P>0 .0 5 )。结论  VEGF可能是诱导大肠癌血管生成的一个重要因子 ,对大肠癌的进展有促进作用 ;p5 3蛋白表达对大肠癌的血管生成及 VEGF表达无明显调节作用。 Objective To investigate the relationship of va scular endothelial grow th factor(VEGF) and p53 protein expression to the angiogenesis in colorectal car cinoma. Methods The expression of VEGF,p53 and microvessel dens ity (MVD) were st udied with LSAB immunohistochemical technique.Results VEGF and MVD positively correlated with Dukes stage (P<0.05),lymph node matastasis (P<0.05),but their relations to tumor histologic type and histologi c grade were not significant (P>0.05).No correlation was found between p53 expression and the clinicopahologic factors above (P >0.05).The MVDs in the VEGF-positive tumors and the VEGF-negative tumors were 24.44±10.34 and 15.88±6.41 respectively,and the diffenerce between them was s ignificant (P<0.01),but no correlations were found between p53 expression and VEGF or MVD(P>0.05).Conclusion VEGF is an impor tant angiogenic factor in colorectal carcinoma and promotes its progression.p53 expression may not be directly involved in the regulation of VEGF expression or angiogenesis in colorectal carcinoma.
