摘要
目的 :探讨CD4 0 及CD4 0 配体 (CD4 0L)分子在活跃期SLE患者外周血单个核细胞 (PBMC)上的表达及在SLE发病中的可能作用机制。方法 :采用流式细胞仪检测 13例初发活跃期及 13例复发活跃期SLE患者PBMC表面CD4 0 及CD4 0L分子的表达。结果 :初发及复发活跃期SLE患者PBMC表达CD4 0 分子 (2 5 .94± 10 .10 ) %及 (2 7.34±12 .94) % ,均较正常对照组 (13.99± 5 .2 6 ) %明显升高 (P <0 .0 0 1) ;初发活跃期SLE患者PBMC表达CD4 0L分子 (1.74±1.6 6 ) %较正常对照组 (0 .91± 0 .39) %亦明显升高 (P <0 .0 5 )。初发与复发活跃期SLE患者之间比较PBMC表达CD4 0及CD4 0L分子无显著性差异。结论 :活跃期SLE患者外周血PBMC存在CD4 0 及CD4 0L分子的异常表达 ,提示CD4 0 及CD4 0L分子在SLE患者T、B细胞之间的信号传导及SLE发病机制中可能起着重要的作用。
Objective:To investigate the expression of CD 40 and CD 40 ligand (CD 40L ) on peripheral blood mononuclear cells (PBMC) in patients with systemic lupus erythematosus (SLE) and its role in the pathogenesis of SLE.Methods:The expression of CD 40 and CD 40L on PBMCs in 13 patients with incipient active SLE and 13 patients with relapsed active SLE was determined by flow cytometry.Results:The expression of CD 40 on PBMCs in both groups of SLE patients [(25.94± 10.10 )% for incipient active SLE;(27.34±12.94)% for relapsed active SLE] was significantly higher than that in normal controls(13.99±5.26)%(P<0.001).The expression of CD 40L on PBMCs in patients with incipient active SLE(1.74± 1.66 )% also increased significantly in comparison with normal controls(0.91±0.39%;P<0.05).There were no statistical differences in the expression of CD 40 and CD 40L on PBMCs between the two groups of patients with active SLE.Conclusion:The abnormal expression of CD 40 and CD 40L on PBMCs in patients with active SLE suggests a disturbed CD 40 -CD 40L signal pathway in SLE patients,which may play an important role in the pathogenesis of SLE.
