摘要
为观察幼猪肺缺血—再灌注过程中肺静脉血肿瘤坏死因子 (TNF -α)含量的变化 ,探讨外源性肺表面活性物质(PS)对缺血—再灌注肺损伤的保护作用及可能机理 ,取 45 ± 天龄幼猪 12只 ,随机分成两组 ( n =6 ) :对照组和PS组 ,对照组阻断前向右肺注入生理盐水 3ml,PS组注入天然PS 40mg/kg ,于阻断右肺动脉前、阻断后 6 0、90min(开放 )、开放后 6 0、90、12 0min等时刻点分别抽取左、右肺静脉血 ,用ELISA法检测TNF -α含量。结果显示 :PS组右肺静脉血中TNF -α含量明显低于对照组 (P <0 .0 1)。
In order to investigate the protective function and mechanism of exogenous pulmonary surfactant (PS) on ischemic-reperfusion injury, the amount changes of tumor necrosis factor-α(TNF-α) in pulmonary vein were observed during the ischemic-reperfusion in young pigs. 12 young pigs (about 45 ± days) were randomizedly divided into two groups: the control group and the PS group, both n=6, the control group were infused with 3ml saline water into the right pulmonary artery before clamping, and the PS group were infused with natural PS 40mg/kg. The blood of left and right pulmonary veins were extracted at the time point before clamping, 60 minute, 90 minute after clamping(opening) and 60 minute, 90 minute and 120 minute after the opening, and the amount of TNF-α were detected by using ELISA method. Results showed that the amount of TNF-α in the right pulmonary vein in the PS group were lower than that in the control group (P<0.01). This study indicated that PS had protective function on ischemic- reperfusion injury.
