高效液相色谱-蒸发光散射检测器法测定保元汤中黄芪甲苷的含量 被引量:7

摘要 目的 :研究保元汤中黄芪甲苷的高效液相色谱 ( HPL C)测定方法。方法 :采用一种新的检测器—蒸发光散射检测器( EL SD)以黄芪甲苷为对照品对保元汤中的黄芪甲苷进行 HPL C分析 ,色谱柱 :Elite-ODS,流动相 :乙腈 -水 -四氢呋喃 ( 33∶ 6 7∶ 4) ;流速 :1m l· m in- 1 ;EL SD参数 :漂移管温度 :10 5℃ ,N2 流速 :2 .82 L· m in- 1。结果 :按选定的色谱条件进行测定 ,平均加样回收率达 97.90 %,RSD为 1.5 1%。样品制备方法的研究结果发现 :黄芪适当粉碎、煎煮 3次 ( 1h/次 ) ,黄芪甲苷的煎出率明显高于不粉碎或提取时间少者。结论 :本文所建立的 HPLC法简便 ,可靠 ,易行 ,可用于保元汤中黄芪甲苷的测定。样品制备方法的研究结果为“黄芪久煎可利于其药效的发挥”的传统观念提供了实验依据。 Objective:To determine astragaloside IV in Bao-yuan Decoction by HPLC.Methods:HPLC and a new detector (ELSD) were used in the quantitative analysis of astragaloside IV in Bao-yuan Decoction by using Ellite-ODS as chromatography column, and acetonitrile:water:tetrahydrofuran (33∶67∶4) as mobile phase. The flow rate of mobile phase was 1.0 ml/min. The tube temperature of the detector was 105℃, the flow rate of N 2 was 2.82 L/min. Result:The average recovery was 97.90% and RSD was 1.51%.The results also showed that powder of Radix Astragali could make much more astragaloside IV than the same amount of the piece, and three times of decoration (one hour each time) made more astragaloside IV than one time or two times.Conclusion:The method was simple, reliable, and was a good method of determination of astragaloside IV in Bao-yuan Decoction. The results of sample preparation also gives an evidence to the traditional opinion that long time decoration is good for effect of Radix Astragali.
