摘要
用酪氨酸吸收系数E法测定多酶片中胃蛋白酶与胰蛋白酶的含量。用不同型号的紫外分光光度计测定不同浓度的酪氨酸吸收度 ,计算出E值 ,用E值法测定胃蛋白酶和胰蛋白酶的活力单位数U。测定胃蛋白酶时 ,可采用酪氨酸E1%1cm .2 75nm =76 .6 (0 .0 6 5mol/L盐酸溶液 ) ;测定胰蛋白酶时可采用酪氨酸E1%1cm .2 75nm =75 .3(0 .2mol/L盐酸溶液 )。该法结果准确 ,精密度高 ,操作简便 。
Purpose The aim is to study the assay of pepsin and trypsin. Methods E 1% 1cm of some different concentration of tyrosine were determined using different type spectropho tometers .The activity of pepsin and trypsin were calculated according to E 1% 1cm of tyrosine. Results E 1% 1cm. 275nm=76.6(0.065mol/L HCl solution) and E 1% 1cm. 275nm=75.3(0.2mol/L HCl solution) can be used to assay the activity of pepsin and trypsin respectively. ConclusionThe method is easy and accurate, and is suitable to assay the activity of pepsin and trypsin in multienzyme.
