骨髓移植后患儿的免疫状态与细胞内钙离子浓度的关系

摘要 骨髓移植后的病人往往伴有免疫功能缺陷,特别是在移植后早期(6个月内),为探讨骨髓移植后病人T细胞内钙离子浓度与免疫状态的关系,用PMA+Ionomycin以及CD3分子Cross-Linking刺激后,作免疫抗体标记,进行FACS分析,了解T细胞内钙离子浓度及细胞增殖情况。结果发现骨髓移植后早期(6个月内)病人细胞内钙离子与正常儿童及移植后18个月病人比较显著升高,同时细胞表面标记检查提示CD45RA^+ T细胞减少,CD45RO^+ T细胞增多,DNA合成明显低下,细胞增殖能力降低。骨髓移植后早期细胞内钙离子浓度异常升高与T细胞功能低下的关系尚待进一步探讨。 It is common that patients have immunodefiency after transplantation of bone marrow (TBM), particularly within the early 6 months after TBM. To explore the relationship between immune state and cytoplasmic free calcium level of peripheral T lymphocyte in children after TBM. FACS analysis was undertaken in 14 normal children and 11 cases who had received TBM, respectively. Using cross-linking PMA + lonomycin and CD3 molecular to stimulate T lymphocyte, corresponding antibody labeling was then performed for the observation of lymphocyte proliferation and cytoplasmic free calcium. The results showed that cytoplasmic free calcium in peripheral T lymphocyte was significantly higher in the children within 6 months after TBM than those in either normal cases or sick children over 18 months after TBM. Besides, the results of the surface marker indicated that CD45RA+ T lymphocyte decreased and CD45RO+ T lymphocyte increased with the reduction of both DNA synthesis and lymphocyte proliferation. Further exploration for the relationship between decreased function and abnormal elevation of cytoplasmic free calcium in T lymphocyte during the early stage after TBM will be required.
