摘要
目的 观察胆管癌细胞内氧自由基对P53蛋白过度表达的影响 ,探索胆管癌细胞内氧自由基与肿瘤病因学的关系。方法 通过细胞培养 ,采用紫外可见分光光度仪等先进设备检测体外培养胆管癌细胞QB中氧自由基中间产物MDA含量 ,应用免疫组化技术 ,检测不同浓度的氧自由基对QB细胞P53蛋白过度表达的影响。结果 胆管癌细胞中P53蛋白过度表达呈阳性。高浓度的氧自由基可明显增加胆管癌细胞中P53蛋白的表达。结论 胆管癌细胞内氧自由基的水平可影响胆管癌细胞P53蛋白表达。
Objective To examine the effect of oxygen free radical on P53 protein overexpression in cholangiocarcinoma cell and study on the relationship between oxygen free radical and tumor pathogenesis. Methods Content of oxygen free radical in cholangiocarcinoma cell were measured with ultraviolet visible light spectrometer and the effect of different concentration oxygen free radical on P53 protein overexpression were detected by immuno histochemical method. Results P53 protein overexpression was seen in cholangiocarcinoma cell. High level of oxygen free radical in the cholangiocarcinoma cell promoted obviously P53 protein overexpression. Conclusion The level of oxygen free radical may play a role on the tumor p53 gene expression.
