摘要
目的 应用荧光偏振法观察 65例 2型糖尿病患者中性粒细胞 (PMN)膜流动性的改变 ,初步探讨这种改变与糖尿病视网膜病变 (DR)的关系。方法 65例无酮症及感染的 2型糖尿病患者 ,3 0例性别、年龄、体重指数 (BMI)相匹配的正常人为对照 ,以DPH为探针测定其PMN的荧光偏振值ρ ,并计算相应的细胞膜流动性 (MF) ,观察糖尿病患者视网膜病变情况 ,探讨PMN的荧光偏振值与糖尿病视网膜病变严重度的关系。结果 ① 2型糖尿病患者的PMN荧光偏振值显著高于对照组 (P <0 .0 1 ) ,PMNMF明显低于对照组 (P <0 .0 5)。②PMN的MF与糖尿病视网膜病变的严重度呈显著负相关 (r=- 0 .3 970 ,P <0 .0 1 )。结论 2型糖尿病患者PMN的MF下降水平与DR的严重度密切相关 ,提示白细胞膜的物理性质改变在DR的发生中有非常重要的作用。
Objective To investigate the alteration of the polymorphonuclear leukocytes (PMN) membrane fluidity(MF) in diabetic patients and the relationship between this alteration and diabetic retinopathy (DR). Methods A total of 65 cases of type 2 diabetic patients without infection and ketoacidosis and 30 cases of healthy controls matched in sex, age and bodymass index (BMI) were chosen for this study and the fluorescence polarization value (ρ)was determined. DR was also observed in diabetic subjects. The alteration and the relationship between PMN, MF and DR were studied. Results The mean ρ value in PMN from diabetic groups was significantly greater than that in PMN from control subjects ( P <0.01); The MF from diabetic group was decreased ( P <0.05). MF of PMN correlated inversely with the severity of DR in type 2 diabetic patients ( r = -0.3970 , P <0.01). Conclusion This study showed that ① Membrane fluidity in PMN from the group of type 2 diabetic patients is decreased. ②There is a significan negative correlation between MF in PMN and severity of DR in type 2 diabetic patients. Alteration of membrane dynamic properties in PMN may play an important role in the occurrance of diabetic microangiopathy and complications in diabetic subjects.
