一氧化氮合酶表达与肝癌发生发展的关系 被引量:1

摘要 目的研究一氧化氮合酶(NOS)与肝癌发生发展的关系。方法用免疫组化法对肝癌及癌旁组织中的3种NOS(诱导型、神经型和内皮型)及血管内皮细胞生长因子(VEGF)的表达进行原位检测和观察。结果非癌组织诱导型NOS多呈阴性或弥漫弱阳性,但部分非癌组织中可见诱导型NOS阳性细胞呈点状分布;癌旁组织多呈阳性;癌组织核心多呈阴性或弥漫弱阳性,但分化中和差的癌组织核心分别有1例NOS呈强阳性;周边癌组织呈局灶阳性,侵入纤维组织中的弥漫癌细胞呈强阳性。VEGF、神经型NOS的分布与诱导型NOS相似。内皮型NOS主要分布在肝癌组织小血管壁内皮及其肌层组织。结论NOS表达与肝组织癌变及肝癌浸润能力有关,NOS表达增加是肿瘤形成中的关键步骤,与获得血管形成和转移表型有关。 Objective To investigatetheeffectof nitricoxideon thedevelopmentandprogressionof hepatocellularcarcinoma(HCC).Methods Theexpressionandlocalizationof vascularendothelialgrowthfactor(VEGF),endothelial,neuronaland induciblenitricoxidesynthases(eNOS,nNOSand iNOS)in the21patientswithHCCwereobservedby immunohisto-chemistry.Results StrongiNOSexpressionwasobservedinthecirrhoticandchronicallyinflammatorylivertissuesadjacent to thetumormargin,whilebeyondtherange1.5cm fromthetumormargin,itsexpressionbecamelightor evenundetectable.MarkediNOSexpressionwas notedon themarginof thetumorsandin HCCcellsthatinvadedthestroma,butin thecore regionof mostof thetumors,onlylightor evenno iNOSexpressionwas detected.Theexpressionsof VEGFandnNOSwere similarto thatof iNOS.eNOS was detectedmainlyin the endotheliumandsmoothmusclecellsof thetumorvessels.Conclusion Excessiveexpressionof NOSmaycontributetothepathogenesisandinvasionof HCC.
