摘要
目的 观察大气压、中压力、高压力培养对离体人脐静脉内皮细胞(Human umbilical vein endothelial cells,HUVECs)凋亡的影响,及血管紧张素转换酶抑制剂卡托普利(Captopril,Cap)对HUVECs凋亡的作用。方法 采用改良的Jaffe’s法,取4-6代HUVECs制成单细胞悬液。采用大气压(OmmHg)、中压力(120mmHg)、高压力(180mmHg)三种条件培养,分别用形态学、DNA凝胶电泳、流式细胞仪等观察各组培养16b、24h、48h HUVECs凋亡情况。结果 与大气压培养比较,中压力培养未见HUVECs凋亡,高压力培养凋亡百分率增加,48h达到最高(P<0.001)。两种浓度Cap均能抑制高压力培养对HUVECs的促凋亡作用。结论 高压力培养能诱导HUVECs凋亡,并呈时间依赖性。而Cap能减轻高压力培养对HUVECs的促凋亡作用。
Objective To clarify whether ambient pressure of culture media affect apoptosis of cultured human umbilical vein endothelial cells, and to observe the effects of captopril on apoptosis. Methods Cells were cultured by modified Jaffe's method. Experiments were carried out using cells from the 4th to the 6th passages. HUVECs were exposed to either atmosphere (0mmHg) , middle pressure(120mmHg) , or high pressure(180mmHg) condition. The changes of apoptosis of HUVECs under different pressure conditions were investigated by morphology, agarose gel electrophoresis and flow cytometric analysis. Results Compared with atmosphere condition,middle pressure condition didn't induce apoptosis of HUVECs, however, apoptosis cell was markedly increased under high pressure condition(P<0. 001). A decrease in apoptosis cells were observed in captopril (10-6,10-5mol/L) treated HUVECs by a dose-dependence(P<0. 001). Conclusions High pressure condition can induce cultured HUVECs apoptosis by a time-ependence. Captopril can inhibit apoptosis of cultured HUVECs under high pressure condition.
