摘要
目的 观察大脑中动脉 ( MCA)缺血后血影蛋白的动态变化 ,寻找反映脑缺血程度的形态学依据 ,并评价尼莫地平的脑保护作用。方法 制作大鼠 MCA局灶性脑缺血模型 ,免疫组化法检测血影蛋白含量 ,缺血后10 min、3 0 min、3 h、6h和 4 8h观察血影蛋白动态变化。结果 缺血后 10 min缺血区域血影蛋白表达降低 ,2 4 h达高峰。尼莫地平可减少缺血区神经元血影蛋白的降解 ,具有脑保护作用。结论 检测血影蛋白可反映神经元缺血损伤后的早期形态学改变。钙离子拮抗剂—尼莫地平有重要的脑保护作用。
Objective In this study we observed the dynamic change of spectrin following middle cerebral artery(MCA) ischemia to search for a morphological basis to reflect the degree of ischemic damage and evaluated the neuroprotective effect of nimodipine. Methods The focal cerebral ischemia model of rat was made with thread embolism of middle cerebral artery. Spectrin was measured by immuno histochemical method. The dynamic change of spectrin were determined at 10min,30min,3h,6h,48h following middle cerebral artery(MCA) ischemia. Results There was a breakdown of spectrin in ischemic region at ten minutes after focal cerebral ischemia,and peaking at 24hr after cerebral ischemia and Nimodipine could diminish the degradation of spectrin and have the neuroprotective effect. Conclusion The measurement of spectrin may provide a method for earlty morphology change of neuron damage after ischemia. Nimodipine,a calcium-channel antagonist has importment brain protection role.
