Study on Culture Techniques of Mature Embryo of Cold Region Rice

试验表明 ,寒地粳稻成熟种胚愈伤组织诱导率很高 ,但分化率不够理想 ,通过调整分化培养基中激素的浓度和配比来提高分化率 ,收到了一定成效。诱导培养基用 6%浓度的蔗糖诱导出的愈伤组织分化率高于 3%和 5 %两种浓度。R2 代存在大量可遗传变异 ,通过田间细致观察比较和选择 ,已获得一批优良变异株系 。

Using nine varieties of japanica rice as materials, culture techniques of mature embryo of cold region rice are studied. The results show that the rate of induction is high,but the rate of differentiation is low.Differentiation can be boosted through various regulator (IAA and KT)concentrations in differentiating medium. 6% sucrose is better than 3% and 5% for differentiation in callus formation medium. There are lots of variation that could be inheried in R 2. Some good materials are obtained through selection, which are in test and planting.
