摘要
探讨了小鼠胚胎成纤维细胞饲养层及干细胞因子 ( SCF)、白血病抑制因子 ( LIF)、碱性成纤维细胞生长因子 ( b FGF)等对体外培养小鼠精原干细胞的作用。结果显示 ,在饲养层上 ,精原干细胞贴壁时间缩至 8~1 2 h,饲养层具有促进其分裂增殖的作用 ;培养液中加入不同浓度的 SCF、LIF及 b FGF,可延长精原干细胞在体外的存活时间 ,其中加入 30μg/L SCF后 ,其体外存活时间与对照组相比差异显著 ( P <0 .0 5 )。
The effects of several factors,such as feeder layer made of mouse embryonic fibrob- last,SCF,LIF,b FGF on mouse spermatogonial stem cells cultured in vitro were investigated. The results showed the anchoring time of these cells could be shortened to8-1 2 h on the feeder layer and the feeder layercould promote proliferation of spermatogonial stem cells.When SCF,L IF and b FGF was added to the medium with variousconcentrations respectively,the average surviving time of sper- matogonial stem cells could be prolonged. And the surviving time of 3 0μg/ L SCF group was signifi- cantly longer than thatof control group ( P <0 .0 5) .
