幼鼠早期癫痫持续状态与成鼠后海马硬化关系的研究 被引量:2

摘要 目的探讨幼鼠早期癫痫持续状态与成鼠后海马硬化之间的关系.方法健康生后15~20天的Wistar幼鼠54只,随机分为生理盐水对照组、地西泮干预组和实验组,采用锂-匹罗卡品腹腔注射制成幼鼠癫痫持续状态模型,并持续追踪至成鼠阶段,进行海马结构的常规病理、超微结构以及Timm组织化学染色观察.结果一些幼鼠早期癫痫持续状态引起的海马结构损伤可持续到成鼠阶段,CA3区苔藓纤维发芽现象较幼鼠更为明显,一些致痫幼鼠发育至成鼠阶段后未见到海马结构的损伤性改变.生理盐水对照组和地西泮干预组未见有海马结构的病理性改变.结论幼鼠早期痫性发作造成的海马结构损伤性改变可持续至成鼠阶段;幼鼠早期长时间的痫性发作与成鼠后海马硬化有关.一些致痫幼鼠发育至成鼠后并无海马硬化现象,说明海马硬化的形成可能与个体的遗传易感性有关. ve To study the relationship between status epilepticus in early naive rats and hipp-ocampal sclerosis of mature rats. Method Healthy postnatal Wistar rat pups (n =54) were divided into 3 groups randomly: control group of normal saline, interference group of diazepam. We used the lithium-pilo-capine induced the model of status epilepticus, after the kindling, we continue to follow up to the stage of nature rats. Morphological changes of hippocampal formation were observed by conventional histopathologic method and transmission electron microscope respectively. In addition, Timm histochemical technique was adopted to study the mossy fiber sprouting in hippocampal formation. Results The neuron loss of hippocampal formation in some naive rats of status epilepticus can last to the stage of mature; Mossy fiber sprouting in hippocampal formation was more obvious than immature rats; No significant pathological changes were found in both the normal saline and diazepam interference groups. Conclusion The damage of hippocampal formation caused by seizures during darly immature rats can sustain to the period of mature rats; The prolonged seizure in early naive rats was related with the hippocampal sclerosis of mature rats; In some of the kindling naive rats, no hippocampal sclerosis were found during the period of mature; It was showed that the hippocampal sclerosis may be associated with an individual genetic susceptibility.
