摘要
目的 通过测定细胞因子IL 6在卵巢癌中的表达、泰素诱导下体外培养卵巢癌细胞局部环境中细胞因子IL 8、IL 6的表达 ,以探求细胞因子IL 6、IL 8在卵巢上皮癌中的临床价值。方法 ①应用MTT比色测定法对 30例卵巢癌患者血清、腹水中细胞因子IL 6进行生物活性测定 ;②应用ELISA方法定量测定 5个卵巢癌细胞系及 10例初代培养卵巢癌细胞局部环境中细胞因子IL 8在泰素作用后水平的变化。结果 ①腹水中IL 6水平明显高于血清中IL 6水平 (P <0 .0 0 0 1) ;②腹水或血清中IL 6水平与卵巢癌分期呈正相关 ;③在 5个卵巢癌细胞系中 ,泰素选择性诱导 3个细胞系IL 8水平显著增高 ,IL 6水平无明显变化 ;在 10例初代培养卵巢癌细胞中 ,5例IL 8水平显著增高。结论 ①腹水或血清中IL 6生物活性的测定有可能成为卵巢癌疾病状态的评估及选择化疗药物的根据 ;②泰素可以选择性诱导IL 8的释放 。
Objective To study the clinical evaluation of IL 6 in ovarian epithelial carcinoma and to investigate the expression of cytokine IL 8 in a series of cell lines and recent explants of human ovarian carcinoma. Methods ①With 7TD1 cell lines,MTT colorimetric method was applied to assay IL 6 activity in serum and ascites of ovarian carcinoma patients;②ELISA was used to determine the levels of cytokines.Results ①IL 6 activity in ascites was significantly greater than that in serum;②IL 6 activity in ascites or serum positively correlated with stages of ovarian carcinoma;③Three of the five cell lines and five of ten samples responded to paclitaxel stimulated with a elevation of ng/ml levels IL 8 protein,but levels of IL 6 was no variations. Conclusion ①The assay of IL 6 activity in ascites or serum is expected to offer help for evaluating patients and selecting chemical drugs in ovarian epithelial carcinoma;②The induction of IL 8 in both proliferating and confluent cultures suggests that paclitaxel responsiveness may depend on direct tumor destruction as well as modification of the local environment the release of IL 8, cytokine with proinflammatory and growth modulating properties.
