MMP-2动态变化在球囊成形术后再狭窄形成中的作用

Time course of MMP-2 in restenosis after balloon angioplasty

目的 探讨基质金属蛋白酶 2 (MMP 2 )在球囊成形术后再狭窄形成中的作用。方法 采用兔的两次损伤髂动脉再狭窄模型 ,应用MMP 2寡核苷酸探针进行原位杂交 ,观察MMP 2在球囊成形术后不同时间点的含量变化。结果 MMP 2在动脉粥样硬化组 (AS)表达为“ +” ,球囊成形术后第 1天组为“ +～ ++” ,第 2天组为“ ++” ,第 4天组为“ +++” ,第 1周组与第 2周组为“ ++” ,第 4周组与第 12周组为“ +～ ++”。结论 MMP 2在球囊成形术后第 1周内表达由弱到强又渐弱 (第 4天为高峰 ) ,发挥其降解Ⅳ型基底膜胶原的作用 ,在再狭窄早期平滑肌细胞的迁移与增殖中起重要作用 ;而 1周后表达的减弱与再狭窄病变中期。

Objective To discuss the effect of MMP 2 (matrix metalloproteinase-2) on restenosis after balloon angioplasty. Methods Using the double injury iliac artery model of the rabbits ,we assessed the time course of mRNA of MMP 2 after balloon angioplasty with situ hybridization methods.Results In atherosclerotic group ,the expression of MMP 2 was '+'; after balloon angioplasty,it was '+'on the first day,'++'on the second day,'+++'on the fourth day.'++'from the first week to the second week.'+～++'after the fourth week until the study was finished.Conclusion The expression of MMP 2 gradually strengthen and become the strongest on the fourth day and then weaken within the first week after balloon angioplasty. The function of MMP 2 is to degrade collagenⅣ of basement membrance,which play an important role in migration and reproduction of smooth muscle cell in the early stage of restenosis. The weak expression of MMP 2 after the first week is concerned with the accumulation of extracellular matrix-collagen and vascular remodeling in the middle and late stage.
