摘要
目的 研究丙型肝炎病毒(HCV)核酸疫苗NV-HC/NS3接种BALB/c 小鼠后,对相应靶抗原攻击的特异性免疫预防和治疗效应、方法以HCV核酸疫苗NV-HC/NS3接种BALB/c小鼠,分别在疫苗接种前后种植能表达靶抗原的BALB/c 小鼠骨髓瘤细胞株SP2/0,观察疫苗接种对肿瘤生长及小鼠寿命的影响。结果种植肿瘤前先接种NV-HC/NS3的小鼠,荷瘤阻抑率达40%;长出肿瘤的小鼠与未接种疫苗的对照组小鼠相比,生存时间延长,生存率提高。先荷瘤后免疫的小鼠,核酸疫苗的接种则能提高小鼠的生存率。结论NV-HC/NS3能诱导小鼠产生针对靶抗原的特异性免疫应答,发挥预防甚至治疗效应。
Objective To investigate the immunoprophylactic and immunotherapeutic effects of DNA vaccine (NV-HC/NS3) after inoculation of mice by target tumor loading technique. Methods Before or after inoculation with N'VHC/NS3 intramuscularly, BALB/c mice in the experimental group were planted with target tumor cells (SP2/0-NS3). which were derived from BALB/c mice and stably transfected with the recombinant plasmid NV-HC/NS3. Tumor growth and survival rate of the mice immunized with NV-HC/NS3 were compared with those in the control group. Results Tumor mass grew well in all mice in the control group in a few days, while no visible tumor mass growth in 40% face immunized with NV-HC/NS3 could be seen. The survival rate and life span of mice in the experimental group were significantly longer than those in the control group. Conclusions Specific immune activity could be induced by immunization with NV-HC/NS3. Such effects are immunoprophylactic and immunotherapeutic.
