摘要
目的 :研究钝顶螺旋藻 (Spirulinaplatensis ,S .P)对硒的富集和有机化作用 .方法 :培养基中添加无机硒亚硒酸钠培养S .P ,2 ,3-二氨基萘 (DAN)荧光分光光度法测定不同增殖期藻体含硒量 .结果 :①S .P生长的迟缓期富集硒较快 ,而在快速期则有机硒增量较明显 ;②在高浓度无机硒条件下培养 ,藻体的总硒、有机硒、有机化率均呈现下降趋势 ;③S .P对硒的生物有机化率在培养第 3d时较低 ,第 6d时较高 ;添加 15 0mg·L- 1硒培养的富硒螺旋藻 ,第 6d时硒主要与水溶性蛋白质结合 ,占 65 .5 %.结论 :S .P中总硒含量、有机硒含量、硒的有机化率及硒在藻体中的分布均呈现动态变化 .
Aim: Dynamics of enrichment and bio-organization of Se in Spirulina platensis(S.P) were investigated. Methods: S.P was cultivated with inorganic Se (Na 2SeO 3), and Se contents in S.P were determined by 2,3-DAN fluorimetry. Results: ① Se was accumulated more quickly in slow phases but organic Se was enhanced more obviously in rapid phases. ② Total Se, organic Se and the rate of Se bio-organization all showed a declining trend in S.P cultivated with high concentrations of inorganic Se. ③ The rate of Se bio-organization was lower at the 3rd day but higher at the 6th day. Above 65% of the total Se was bound to water-soluble proteins at the 6th day in the reproduction phases of S.P. Conclusion: The contents of total Se, organic Se, rate of Se bio-organization and the distribution of Se in S.P all showed dynamical changes in different reproduction phases.
