甘肃回汉族人群D1S80位点扩增片段长度多态性研究 被引量:2

Genetic Studies on the Variable Number of Tandem Repeat Locus D1S80 in Hui and Han Ethnic Groups of Northwest China

摘要 报道了对甘肃省 10 8名汉族和 72名回族无关个体 D1S80位点扩增片段长度多态性的等位基因频率调查分析 .已在汉族中发现 2 1个等位基因 ,71种基因型 ,基因频率为 0 .0 0 5~ 0 .111,杂合度为 82 % ,个体识别率为 0 .95 ,在回族人群中发现 2 2个等位基因 ,5 1种基因型 ,基因频率为0 .0 0 7~ 0 .15 3,杂合度为 86 % ,个体识别率为 0 .92 ,基因频率符合 Hardy- Weinberg法则 .结果提示甘肃省回族和汉族之间在其等位基因数目及频率分布上均存在显著性差异 ,进一步的分析还表明甘肃省汉族人属于北方群体 ,支持了有关中国大陆存在南北两大群体的观点 . The Variable Number of Tandem Repeat (VNTR) locus D1S80 is one of the valuable genetic markers for studying population genetics, forensic identity, paternity testing and medical genetics. The genetic polymorphism of VNTR locus D1S80 of Hui and Han ethnic groups in Gansu Province was studied by using the amplified fragment length polymorphism (Amp FLP) technique. Out of 108 unrelated Han individuals, a total 21 different alleles could be distinguished. The alleles ranged from 0.005 to 0.111 in frequency, with an observed heterozygosity of 82%. And out of 72 unrelated Hui individuals, a total 22 different alleles were observed, which varied from 0.007 to 0.153 in frequency and showed a heterozygosity of 86%. A x  test demonstrated that the allele frequency distributions of both samples were consistent with the Hardy Weinberg equilibrium. There was a significant difference in the number of D1S80 VNTR locus and allele frequencies of population between Huis and Hans. A further analysis indicated that Hans in Gansu should belong to North China population. This conclusion supports the view that the Chinese nation is composed of two major populations, i.e. northern and southern ones.
