摘要
目的探讨免疫磁珠法(MiniMACS)能否用于纯化小鼠骨髓CD34+造血干细胞。方法应用免疫磁珠纯化小鼠骨髓CD34+细胞,流式细胞仪(FACS)评估纯化效率,检测纯化后的细胞活力。结果纯化后CD34+细胞纯度81.5%(范围76.80%~85.38%),回收率47.54%(范围40.50%~54.31%),纯化前后细胞活力不受影响(P=0.169)。结论应用MiniMACS纯化小鼠骨髓可获得高纯度CD34+细胞,并不影响细胞活力。
Objective To assesstheefficiencyof immunomagneticbeadsinthepurificationof CD34 + cellsfrommurinebone marrow.Methods CD34 + cellswereisolatedfrommurinebonemarrowusingimmunomagneticbeadson MiniMACSdevice.Fluorescenceactivatedcellsorter(FACS)was employedto determinethepercentagesof CD34 + cellsbeforeandafterthe purificationproceduresfollowedby calculationof theviablecellssurvivingthepurificationby meansof trypanbluestaining.Results Thepurityof CD34 + cellsamountedto81.5%(rangingfrom76.80%to85.38%)afterthepurificationwitha median recoveryrateof47.54%(rangingfrom40.50%to54.31%).Thecellviabilitywasnotnotably impaired(P=0.169).Conclusion HighlypurifiedCD34 + cellscanbe obtainedfrommurinebonemarrowusingMiniMACSwhichdoesnotimpairtheviability of thederivedcells.
