17-β-雌二醇对氧化低密度脂蛋白诱导内皮细胞生长调节致癌基因α的影响

摘要 目的 研究 17 β 雌二醇对氧化低密度脂蛋白 (oxLDL)诱导内皮细胞产生生长调节致癌基因α(GROα)的影响。方法 以体外培养的人脐静脉内皮细胞 (HUVECs)为模型。将oxLDL与HUVECs孵育 ,及HUVECs经 17 β 雌二醇预处理或 17 β 雌二醇和他莫昔芬共同预处理后 ,再与oxLDL孵育 ,检测内皮细胞表面的GROα蛋白量。结果 oxLDL可明显促进内皮细胞产生GROα(P <0 .0 1) ,17 β 雌二醇预处理抑制了oxLDL诱导的内皮细胞表达的内皮细胞表面GROα蛋白 ( P <0 .0 1) ,而他莫昔芬逆转了 17 β 雌二醇的抑制作用 ( P <0 .0 1)。结论 通过内皮细胞上雌激素受体α,雌激素明显抑制oxLDL诱导血管内皮细胞产生的内皮细胞表面GROα蛋白。 To study the effects of 17 β estradiol on GROα surface expression by endothelial cells incubated with oxLDL.Methods Human umbilical vein endothelial cells (HUVECs) were obtained in vitro culture.HUVECs were incubated with oxLDL before and after HUVECs treated with tamoxifen and (or) 17 β estradiol,and surface expression GROα by endothelial cells were measured with ELISA.Results oxLDL significantly upregulated GROα surface expression by endothelial cells (P<0.01).17 β estradiol could decrease GROα surface expression by endothelial cells incubated with oxLDL(P<0.01).However,tamoxifen reversed the inhibition of GROα surface expression by 17 β estradiol in endothelial cells incubated with oxLDL(P< 0.01 ).Conclusion 17 β estradiol can obviously reverse oxLDL induced GROα surface expression by vascular endothlial cells through the estrogen receptor α.
