HINS-R DNA Ex-17基因多态性与细胞低镁介导的DM高血压高发倾向的关系

摘要 目的 :探讨人胰岛素受体 (HINS R)基因多态性及低镁与DM患者高血压高发倾向的关系。方法 :采用PCR SSCP技术和ICP AES法分析了 6 3例 2型DMHINS RDNAEx 17多态性情况及RBCMg2 + 含量。结果 :Ex 17位点呈现多态性 5例 ,其RBCMg2 + 明显低于其它病例 (P <0 .0 1) ,而FINS水平明显高于对照组 (P <0 .0 1) ,其它DM伴高血压倾向者RBCMg2 + 也低于不伴高血压的DM者 ,血清Mg2 + 两组比较差异无显著性 (P >0 .0 5 )。结论 :HINS RDNAEx 17多态性常与高胰岛素血症或IR关联 ,且常伴细胞内Mg2 + 储备低下的情况 ,可被血清Mg2 + 正常的表面现象所掩盖 。 Objcetive:To study the relation between HINS R gene poymorphism,and intracellular free Magnesium insufficiency and pathogensin of hypertension high attack tendency in patients with diabetes mellitus(DM).Methods:HINS R DNA Ex 17 gene polymorphism and Megnesium level in red blood cell (RBC) were analysed with PCR/SSCP and ICP ARS in 63 cases type Ⅱ DM.Results:There were Exon 17 polymorphism deformity in 5/63 cases,and its RBC Mg 2+ was lower than that of the patients with normal Exon 17 polymorphism(P<0.01).RBC Mg 2+ level in DM patients with hypertension tendency was also lower than that of DM patients without hypertension.FINS level was higher than that of control patients.Conclusion:HINS R DNA Ex 17 polymorphism is related to IR.The patients with Exon 17 poymorphism deformity in type Ⅱ DM are generally associated low RBC Mg 2+ reservetion.Then Exon 17 polymorphism deformity and low RBC Mg 2+ can reinforce hypertension high attack tendency in patients with type Ⅱ DM.
