妊娠肝内胆汁淤积症患者血清对体外培养的脐带静脉内皮细胞ICAM-1、P-selectin表达的影响

The Effect of Sera from Intrahepatic Cholestasis of Pregnancy on the Expressions of ICAM-1 and P-selectin on the HUVEC

目的 探讨妊娠肝内胆汁淤积症 (ICP)患者血清对脐带静脉内皮细胞间粘附分子 - 1(ICAM- 1)、选择素 - P(p- selectin)表达的影响。 方法 采用脐带静脉内皮细胞培养、流式细胞仪检测的方法。 结果 (1)经 ICP患者血清刺激后 ICAM- 1和 P- selectin的表达与未刺激时相比有差异。(P<0 .0 5 )。(2 )经 ICP患者血清刺激后 ,随着血清量的增加、作用时间的延长 ,ICAM- 1的表达逐渐增强 (P<0 .0 5 ) ,而 P- selectin的表达与作用时间无关 (P>0 .0 5 )。 (3)地塞米松可以抑制 ICP血清对新生儿脐带内皮细胞 ICAM- 1、P- selectin的上调作用 (P<0 .0 5 )。 结论 ICP患者血管内皮细胞有一活化过程 ,而导致血清成分的改变。

Objective To study in vitro ICAM 1 and P selectin expressions of endothelial cells treated with sera from intrahepatic cholestasis of pregnancy (ICP). Methods HUVEC were cultured and stimulated with sera, and the expression of ICAM 1 and P selectin by flow cytometry were measured. Results (1) The expressions of ICAM 1 of P selectin on the HUVEC surface after stimulation with sera from women with ICP were higher than those of without stimulation in normal pregnancy ( P <0.05). (2) The expressions of ICAM 1 and P selectin on HUVEC increased when the concentration and action time of sera from women with ICP in the normal pregnancy increased ( P <0.05). (3) Dexamethasone(DEX) significantly attenuated expressions of ICAM 1 and P selectin on HUVEC which were induced by sera from ICP ( P <0.05). Conclusions There is a course of activation in vascular endothelial cells of patients with ICP, which results in the changes of the component of sera. Sera from women with ICP may contain activation factors which induce and increase the expressions of ICAM 1 and P selectin.
