摘要
目的 研究P2 7蛋白在非小细胞肺癌 (NSCLC)组织中表达、降解及其与临床病理特征、生存期之间的关系 ,以明确P2 7蛋白在NSCLC中表达水平、受调控机制及其对判断NSCLC预后的价值。方法 免疫组化研究P2 7蛋白在NSCLC组织中的表达 ;泛素—蛋白酶体降解法检测NSCLC组织中P2 7蛋白降解活性 ;用统计学方法分析P2 7蛋白水平与 86例NSCLC患者术后生存期之间的关系。结果 ①正常肺组织的P2 7蛋白降解活性均低 ,10例NSCLC组织的P2 7蛋白降解活性有不同程度的增强 ,并与P2 7蛋白的细胞阳性率呈密切负相关。② 86例NSCLC组织中 ,P2 7蛋白表达阴性者、细胞阳性率≤ 5 0 %者及>5 0 %者分别占 5 .8%、6 9.8%、2 4.4%。③NSCLC组织中P2 7蛋白表达高低与患者预后有关。P2 7蛋白低表达者 (细胞阳性率≤ 5 0 %及阴性者 )术后平均生存期 ( 32 .8个月 )低于P2 7蛋白高表达者 (细胞阳性率 >5 0 %者 )的术后平均生存期 ( 4 9.6个月 ) (P <0 .0 1)。④经Cox模型多因素分析 ,P2 7蛋白表达高低是影响NSCLC预后的一个独立因素。结论 ①NSCLC组织存在P2 7蛋白表达的降低或缺失。②NSCLC组织比正常肺组织的P2 7蛋白降解活性有不同程度增强 ,并与P2 7蛋白水平呈负相关 ,是影响P2 7蛋白水平的主要因素之一。③P2
Objective By studing the expression and degradation of P27 protein in NSCLC and their correlation with clinical pathologic features and prognosis of NSCLC to elucidate the significance of expression of P27 protein its regulating mechanism and the value in the prognosis of NSCLC.Methods Immunohistochemincal detection of P27 protein in NSCLC; ubiquitin瞤roteasome detection of the P27 protein degradation in NSCLC; statistics method analysis of the relation of the expression of P27 protein to patients of postoperative survival duration.Results ①Compared with normal lung,P27 protein degradation of NSCLC increased to a greater or less extent, and was closely negatively related to the cell of P27 protein.②In 86 cases,patients with negative expression of NSCLC P27 protein accounts for 5.8%,those with positive cell rate <50% and positive cell rate >50% account for 69.8% and 24.4% respectively.③In NSCLC,P27 protein positive rate related to the survival duration and survival rate of the patients of postoperation,the mean survival duration and rate of low P27 protein expression was apparently below that of high P27 protein expression(P<0.01).④Multifactor analyse of Cox model showed that expression of P27 protein was the main related factor which could affect the prognosis of NSCLC.Conclusions ①The expression of P27 protein decreased in NSCLC.②P27 protein degradation activity increased in NSCLC compared with normal lung and was negatively related to the level of P27 protein,which were of the main factors affecting P27 protein level.③Low expression of P27 protein was a marker of poor prognosis of NSCLC.
