摘要
采用制备的兔抗血清建立了甲胺磷 (methamidophos,MTP)残留的间接竞争性ELISA方法 ,证明了抗血清的特异性较好 ,并确立了测定参数 :抗血清与包被抗原的最佳工作浓度分别为 1∶30 0 0和 1∶5 0 0 0 ;最适检测范围为 0 .0 1~ 0 .1μg/ml;批内变异系数 7.5 6 % ,批间变异系数 5 .70 %。
Methamidophos(MTP) is a pesticide of high toxicity and its residue has been threatening the health of human being. Indirect competitive enzyme linked immunosorbent assay(ELISA) was developed for the detection and estimation of methamidophos residue. The resulting antibodies were specific for MTP. The optimum concentration of antiserum and antigen coating were 1∶3?000 and 1∶5?000, respectively. The linear detection ranged from 0.01 μg/ml to 0.1 μg/ml, with mean within and between assay variability of 7.56% and 5.70%, respectively.
